Hi! To travel from Guatemala City to Flores, it takes several hours, from what I know. I can’t say exactly how long because I’ve either flown or started the trip from Belize. But you should plan for several hours.

If your trip is 300 km or less, budget half a day for travel time. If it's more than 300 km, budget a full day.

The journeys are long, but your route is doable by shuttle—these minibuses are direct, unlike local buses, so they’re faster.

Personally, I’d do Lanquín → Flores → Río Dulce, and if I had to skip a stop, it’d be Río Dulce. But you can easily spend several days around Lake Atitlán or in the Petén region.
